Three-dimensional (3D) brain organoids derived from human pluripotent stem cells (hPSCs), including embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs), appear to recapitulate the brain’s 3D cytoarchitectural arrangement and provide new opportunities to explore disease pathogenesis in the human brain. A new three-dimensional map called BigBrain is the most detailed ever constructed, and should lead to a more accurate picture of how the brains different regions function and interact. As a proof-of-concept study, the device is then used in the intensive care unit for online monitoring of a brain injury patient, demonstrating its capabilities for clinical monitoring. 3D models can be saved for later use or screen-captured for slide presentation or printing. The folds, creases and intricate internal structures that make up the human brain are being revealed in unprecedented detail. The device is employed to record simultaneous glutamate, glucose and lactate concentration changes simulating the physiological response to spreading depolarisation events in cerebrospinal fluid dialysate. The time resolution of the device is characterised by recording short lactate concentration pulses. It was created by the Dolan DNA Learning Center of Cold Spring Harbor Laboratory, in collaboration with Vivid Apps and AXS Biomedical Animation Studio. The term MPS in this review refers to any of these models with the focus on 3D brain models or brain organoids ( Lancaster and Knoblich, 2014 Pasca et al., 2015 Di Lullo and Kriegstein, 2017 Koo et al. The optimisation process of the microfluidic channel fabrication, including for different types of 3D printer, is explained and the resulting improvement in sensor response time is quantified.
